Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen so that water becomes ice; then the surrounding pressure is lowered below the vapor pressure of ice. Heat is applied gently so ice changes directly to vapor without passing through a bulk liquid phase. The vapor is collected on a cold condenser, leaving a dry porous matrix. This process differs from simple evaporation because the material remains frozen during the main drying stage.
The process usually has three stages: freezing, primary drying, and secondary drying. Freezing sets the ice crystal structure and can determine pore size in the final cake. Primary drying removes free ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, although some water may remain as part of the solid. Cycle parameters depend on formulation, fill volume, vial type, and equipment performance.
The process generally proceeds in three stages: freezing, primary drying, and secondary drying. During freezing, controlled cooling converts water into ice and may also crystallize or vitrify solutes. In primary drying, the pressure is lowered below the triple point, and heat is supplied so ice sublimes directly to vapor. Secondary drying removes water that remains bound to the solid matrix, yielding a low final water content. Product temperature must stay below the collapse or glass transition temperature to maintain structure. Cycle design therefore balances shelf temperature, chamber pressure, and time.
Freeze-drying is used for materials whose activity or structure depends on low temperature and low water content. Examples include certain biologics, diagnostic reagents, starter cultures, coffee, and porous inorganic precursors. The dried product forms a cake whose porosity aids rapid wetting and dissolution. Main drawbacks are high energy use, long cycle times, and sensitivity to formulation and equipment variation. Questions remain about how freezing rates and ice morphology affect batch uniformity, especially when moving from laboratory to production scale.
| Property | Value | Notes |

| Common synonyms | Freeze-drying, lyophilisation | Lyophilisation is the British spelling; the process is not simple evaporation. |
| Primary drying pressure | 0.05–0.3 mbar | Pressure must remain below the vapor pressure of ice at the product temperature. |
| Sublimation temperature | Below 0 °C | Ice changes directly to vapor while the product remains frozen. |
| Typical shelf temperature | −40 to −10 °C | Exact setting depends on formulation critical temperature and equipment. |
| Cycle duration | 12–72 hours | Time varies with fill volume, formulation, and dryer performance. |
A formulation often contains excipients that protect the active ingredient during freezing and drying. Bulking agents provide structure, while lyoprotectants stabilize sensitive molecules. The freezing step can produce ice crystals whose size and distribution affect the drying rate, and cycle design includes freezing, annealing, and drying phases. If the product temperature rises above a critical value, the cake may collapse or lose its porous structure. Successful lyophilization therefore depends on the interaction between formulation, equipment, and cycle design.
Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen to convert liquid water into ice. Next, the pressure is reduced below the triple point of water so that ice changes directly into vapor without passing through a liquid phase. This step is called primary drying. The result is a porous solid or cake that retains the original shape of the frozen solution.
After primary drying, secondary drying removes water that remains bound to the material. This stage raises the shelf temperature while maintaining low pressure, which encourages desorption of unfrozen water. Residual moisture can be reduced to a low percentage, improving stability for many products. The process parameters, including freezing rate, shelf temperature, and chamber pressure, influence the final pore structure and reconstitution behavior. Control of these variables helps prevent collapse or meltback during drying.
Freeze-drying is distinct from simple evaporation and from spray drying. Evaporation removes water at temperatures above freezing, while spray drying rapidly dries droplets in a heated gas stream. Lyophilization avoids high temperatures, which can be useful for heat-sensitive materials such as proteins, vaccines, and some foods. The porous cake produced by sublimation dissolves or rehydrates more quickly than a dense dried mass. Not all materials tolerate freezing or the pH shifts that can occur as solutes concentrate during ice formation.
Lyophilization removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts liquid water into ice and fixes the structure of the sample. After freezing, primary drying lowers pressure so ice changes directly to vapor without passing through a liquid phase. Secondary drying then removes bound water that remains after ice sublimation. The result is a dry, porous solid that often retains its original shape.
== Treatment == Avoid cold weather. Treat the underlying lymphoma. No cold drinks; all drinks should be at room temperature (or above). Requires heater to maintain temperature in cold places. Treatment with rituximab has been described. The treatment of cold agglutinin disease depends on many factors including the severity of the condition, the signs and symptoms present in each person, and the underlying cause. For example, in those affected by secondary cold agglutinin disease, it is important to diagnose and treat the underlying condition which may include certain types of cancer; bacterial, viral, or parasitic infections; and/or other autoimmune disease. People with few symptoms and/or mild anemia may not require any specific treatment. These cases are often managed by simply avoiding exposure to the cold. In severe cases of hemolysis, medical interventions may be necessary. Rituximab (an antibody that selectively reduces specific types of immune cells) is effective in about 60% of cases of severe cold agglutinin disease. Medical researchers have found the response to rituximab is seen on average within 1 to 2 months of treatment and the effect of the treatment lasts for about 1 to 2 years. Rituximab may be used after a second and even a third relapse, however the success rate is less. Combined treatment with rituximab and fludarabine has resulted in higher response rates (76% of cases) and longer periods of remissions (on average, 6.5 years).
Kyle Hardingham (born 1 September 1988) is a former professional Australian rules footballer who played for the Essendon Football Club in the Australian Football League (AFL). Hardingham was selected by Essendon with the seventh pick in the 2010 AFL Pre-Season Draft. Originally from East Fremantle in the WAFL, he represented Western Australia in the state game against South Australia in 2009. He made his AFL debut against North Melbourne in round 17 of the 2010 AFL season, kicking four goals, helping Essendon achieve an upset victory by 3 points. The following Friday night, he kicked two goals in a win against St Kilda. He managed to kick two goals once again in the next game, a 76-point loss to Carlton, although he also kicked two behinds as part of the wayward kicking by the entire Essendon team, which managed 9.19 for the night. Hardingham was delisted at the conclusion of the 2014 AFL season. He remained with Essendon as a VFL-listed player in its reserves team in 2016. Hardingham, along with 33 other Essendon players, was found guilty of using a banned performance-enhancing substance, thymosin beta-4, as part of Essendon's sports supplements program during the 2012 season. He and his teammates were initially found not guilty in March 2015 by the AFL Anti-Doping Tribunal, but a guilty verdict was returned in January 2016 after an appeal by the World Anti-Doping Agency. He was suspended for two years which, with backdating, ended in November 2016; as a result, he served approximately fourteen months of his suspension and missed the entire 2016 season.
== History == The use of antibodies to treat diseases can be traced all the way back to the late 1800s with the advent of diphtheria antitoxin for the treatment of diphtheria. It wasn't until the 1900s that the newly emerging class of naturally derived medications such as sera, vaccines, and antitoxins began to be referred to as biologics. The definition for biologics and biological therapy has changed a lot since. The development of recombinant DNA technology in the 1970s shaped the modern understanding of what constitutes as biological therapy, which often does not include traditional biological substances like vaccines. Today, biological therapy most commonly refers to the use of proteins, such as monoclonal antibodies, to regulate the immune system in the treatment of disease. In 1975, Georges J. F. Köhler and César Milstein generated the first monoclonal antibodies using their own hybridoma technology. They started the field of monoclonal antibody development and won the Nobel Prize for Medicine in 1984 for their work. Soon after, muromonab-CD3 became the first fully licensed monoclonal antibody in 1986 for its use in treating kidney transplant rejection. Since then, over 70 monoclonal antibodies have been approved by the FDA. The advancements in biological therapy greatly changed how IBD is treated. Patients with Crohn's disease and ulcerative colitis show an increase in proinflammatory cytokines such as IL-1, IL-6, IL-8, IL-23, and TNF. In 1988, a monoclonal antibody called infliximab was discovered at New York University's School of Medicine.

==== Distribution ==== Enobosarm is a small-molecule and highly lipophilic compound. Compounds of this type are typically able to diffuse freely through biological membranes such as cell membranes and barriers like the blood–brain barrier. This is in fact essential for the action of nuclear receptor ligands like enobosarm since their biological targets (the androgen receptor in this case) are located intracellularly. One in silico study predicted that, on the basis of its overall physicochemical properties (but not considering active transport), enobosarm would be unlikely to cross the blood–brain barrier and hence would be a peripherally selective drug with reduced or no central nervous system effects. However, in a rat tissue distribution study, enobosarm was found to be concentrated in brain tissues to a similar extent as other target tissues like skeletal muscle, bone, prostate, and seminal vesicles. This is consistent with enobosarm producing centrally mediated effects in humans like suppression of LH and FSH secretion. Enobosarm does not bind to sex hormone-binding globulin.
==== Bedding ==== Silver is widely studied for its antimicrobial properties, particularly its ability to inhibit microbial growth and reduce odor formation in textiles. It is also used in some textile applications for its thermal conductivity. Bed sheets, bed pillows, comforters, and other bedding products infused with ions are marketed for their antimicrobial and antifungal additives.

=== DNA chips === Early biochips were based on the idea of a DNA microarray, e.g., the GeneChip DNAarray from Affymetrix, which is a piece of glass, plastic or silicon substrate, on which pieces of DNA (probes) are affixed in a microscopic array. Similar to a DNA microarray, a protein array is a miniature array where a multitude of different capture agents, most frequently monoclonal antibodies, are deposited on a chip surface; they are used to determine the presence and/or amount of proteins in biological samples, e.g., blood. A drawback of DNA and protein arrays is that they are neither reconfigurable nor scalable after manufacture. Digital microfluidics has been described as a means for carrying out Digital PCR.

Desmosine is an amino acid found uniquely in elastin, a protein found in connective tissue such as skin, lungs, and elastic arteries. Desmosine is a component of elastin and cross links with its isomer, isodesmosine, giving elasticity to the tissue. Detection of desmosine in urine, plasma or sputum samples can be a marker for elastin breakdown due to high elastase activity related to certain diseases.
== Hybrid techniques == Combinations of the above techniques produce "hybrid" or "hyphenated" techniques. Several examples are in popular use today and new hybrid techniques are under development. Hyphenated separation techniques refer to a combination of two or more techniques to separate chemicals from solutions and detect them. Most often, the other technique is some form of chromatography. Hyphenated techniques are widely used in chemistry and biochemistry. A slash is sometimes used instead of hyphen, especially if the name of one of the methods contains a hyphen itself. Examples of hyphenated techniques:
=== Interactions with Dictyostelium === The slime mold Dictyostelium discoideum contains "farmer" individuals that live in association with Pseudomonas fluorescens. Two strains of the bacterium are associated with the farmer. One strain serves as a food source and the other strain serves as a symbiont that produces beneficial secondary metabolites. The main genetic difference between these two strains is a mutation of the global activator gene called gacA. This gene plays a key role in gene regulation; when this gene is knocked out in the nonfood bacterial strain, it loses its special secondary metabolites and, independently, is transformed into a food strain.
== Personal life == On July 20, 1957, Sunny married Prince Alfred Eduard Friedrich Vincenz Martin Maria Auersperg. He came from a very distinguished Austrian princely family that once ruled over the Principality of Auersperg, but due to the collapse of the Austrian Empire, his family became relatively impoverished. Sunny and Alfred met while he was her tennis instructor in a Swiss resort. They had two children together:
=== AFL policy === The AFL is a signatory to ASADA and WADA, and its policy on performance-enhancing drugs is based upon those organisations. For non-performance-enhancing drugs, the AFL currently has a "three-strikes" policy, under which only the player and the club doctor are aware of any positive tests until a third such test is received. After the third strike, the club is made aware of the situation and the player may face disciplinary action. This policy has faced criticism for leniency from anti-doping bodies, particularly the World Anti-Doping Agency (WADA), and as such has attracted much media scrutiny and public debate.

The main change is sublimation, in which ice becomes water vapor without melting into liquid water. This occurs when the chamber pressure is held below the vapor pressure of ice while mild heat is supplied. The result is a dry, porous solid that retains much of its original shape.
Freezing determines ice crystal size, solute distribution, and the pore network left after drying. A slow or fast freezing rate can produce different cake structures and affect reconstitution. It also sets whether the formulation follows an amorphous or crystalline drying path.
It removes most free water during primary drying and part of the bound water during secondary drying. A small residual moisture content often remains and is specified for each product. Complete removal is generally neither practical nor desirable for stability.
Yes. Lyophilization and freeze-drying are synonyms for the same vacuum-assisted sublimation process. The term lyophilization is more common in pharmaceutical and laboratory settings, while freeze-drying is widely used in food and general contexts.
